It sounds like you or someone else hacked the phone to unlock it. Updating it locked it to the original carrier.
How did you unlock it? The ONLY legitimate way is to ask the carrier it is locked to to unlock it. Any other way requires hacking or jailbreaking the phone.

Your iPhone was hacked or jailbroken before and through the update it locks back to the original carrier. Only the original carrier can unlock the iPhone. You have to contact them and find out what the procedure you needs to follow.

I just purchased an Simfree & UNLOCKED iPhone 5 32GB from an Regent Street - Apple store in UK.
The part number is MD299B/A.
When I try to activate it with my iTunes, is gives the error below and does not seem to activate:
"The SIM card inserted in this iPhone does not appear to be supported. Only compitable SIM cards from a supported Carrier may be used to activate iPhone.Please insert SIM card that came with your iPhone or visit a supported Carrier's store to receive a replacement SIM card."

Those are 2 different things.
I'm guessing Apple Care is correct. The store is incorrect. Get the store on the phone. Then get Apple care on the phone in a conference call. If the store says it should be unlocked, then they need to get that fixed in the database. They apparently made a mistake when they recorded the serial number in their system. -

You purchased a carrier locked iPhone. If you don't intend on using it with the carrier it is locked to, return it for a refund & either purchase an officially unlocked iPhone or one locked to the carrier you want to use.

It's locked to a carrier other than the one you're trying to use. Either sign up for cellular service with the carrier it's locked to, or contact them to see if you can have it unlocked to that you can use the carrier of your choice.

Your phone was previously hacked to get it to work on a carrier other than the one it was locked to. Restoring it restored the carrier lock. Only the carrier it is locked to can authorize an unlock. Not all carriers will do this. You will have to find out who it is locked to and contact them.
