This hasn't happened till this upgrade.
A note at 1 1 1 1, in a region placed at 1 1 1 1, won't play when I start playback from (you guessed it...) 1 1 1 1
Quit Logic, rebooted and opened song, made no difference.
Anyone else?

I've had this for quite some time. I believe It all started when the tool became "Pro" (but I could be wrong). The first midi note on an audio instrument track. I remember Justin C saying that Audio instruments go to sleep, and this can be worked out by adding a non-playing midi event (like a cc) could wake up the audio instrument, but I don't agree with this level of house keeping to just get the first note to play. Nothing against Justin C, he does a great job here!
In my opinion, this is an issue that still needs to be worked out by the development team.
