All iTunes Music, Podcasts, Apps gone

I opened iTunes last night and all my music, podcasts, and apps were completely gone. I logged out of iTunes and same thing when I logged back in. I reinstalled iTunes and same problem. Also, I have an iPhone 4. Please help, I don't know what happened.

Something trashed your itunes database file.
Usually it's security software, or backups software that locks itunes database file and prevents it from being saved correctly.
Are you running IObits or Advanced Care? How about Carbonite? There are literally dozens of programs that could be doing this.
You need to tell whatever software to leave the itunes database file alone.

Same as you would if you bought a pc; copy everything from your old computer to your new one. Then you can just sync everything from the new computer, as you did with the old one.

It won't unless a problem occurs. Back them up, uninstall iTunes and all related components, and reinstall them.

My wife and I have been sharing an iTunes accound and library on our own iPods then iPhones for years without much trouble. Now, Mai ly because of the cloud and a new desktop, I think it's time to create her own account but I don't want to have to repurchase all her music and apps. Is this poosible?

Yes, with the credentials of the account that the media/content was purchased with.
Content is permanently tied to the account it is purchased with and cannot be merged, moved, or transferred to another account.

1) authorize iTunes on the new machine for your iTunes store account(s)
2) connect your device and right-click or control-click it in the iTunes Source list, then choose transfer purchases from the shortcut menu that appears.
3) sync the device
iTunes will wipe the iPhone but, since you transferred your purchases in the previous step, your content will be in your library and you can re-populate the iPhone with it.

I want to transfer all my music, iPhone apps, & movies from iTunes on my desktop computer to iTunes on my laptop computer. Is there any way to do that.

These are two possible approaches that will normally work to move an existing library to a new computer.
Method 1
Backup the library with this User Tip.
Deauthorize the old computer if you no longer want to access protected content on it.
Restore the backup to your new computer using the same tool used to back it up.
Keep your backup up-to-date in future.
Method 2
Connect the two computers to the same network. Share your <User's Music> folder from the old computer and copy the entire iTunes library folder into the <User's Music> folder on the new one. Again, deauthorize the old computer if no longer required.
Both methods should give the new computer a working clone of the library that was on the old one. As far as iTunes is concerned this is still the "home" library for your devices so you shouldn't have any issues with iTunes wanting to erase and reload.
I'd recommend method 1 since it establishes an ongoing backup for your library.
If you have an iOS device that syncs with contact & calendar data on your computer you should migrate this information too. If that isn't possible create a dummy entry of each type in your new profile and iTunes should offer to merge the existing data from the device into the computer, otherwise the danger is that it will wipe the information from the device.
If your media folder has been split out from the main iTunes folder you may need to do some preparatory work to make it easier to move. See make a split library portable.
Should you be in the unfortunate position where you are no longer able to access your original library, or a backup of it, then see Recover your iTunes library from your iPod or iOS device for advice on how to set up your devices with a new library with the maximum preservation of data.

Last night I decided that I would go and update my ipod for the first time to the latest version of the system. I completely forgot that I still had not backed up any of my purchased music and apps and of course when the update was finished, I found that all my music and apps had been erased.
