I have an iphone 3gs, and i updated a latest version 6.01, and now when i updated that, i can't connect my iphone to iTunes, and activate it. what should i do

This happens to iPhone which is unofficially unlocked. Nobody here can tell you the carrier to which your iPhone was locked.

Was the phone hacked or jailbroken to use it with a different carrier than the one it was locked to? If it was then it is again locked to that carrier, and there is nothing you can do about it that can be discussed in an Apple hosted forum.

Well, it's a hacked and the update locks it back to the original carrier. If officially unlocked, updates will not lock it back.
Do you know only the carrier can officially unlock iPhone, not even Apple can do that let alone some website?

Basically, Before i updated it worked fine, Now i have updated it just comes up no service. I have downgraded it back to 4.1. but now cant update it to 5.1.1 again as i keep getting the 3194 error on itunes. Will there be a fix for ios6 to sort the no service issue out?

You cannot do anything without a sim for the carrier the iPhone is locked to. With that you can attempt to restore using the recovery mode described in http://support.apple.com/kb/HT1212 but note that if the previous owner installed an Apple ID and password and turned on Settings > iCloud > Find My Phone, you will never get it to work without the Apple ID and password. That is the Activation Lock, an anti-theft protection system.

Can't Synq iPhone 3GS Calendars after installing iOS6, any suggestions?
I'm all mac-based and followed the instructions for the upgrade to iOS6 software for iPhone but now my calendars won't synq. When I connect to iTunes and run the synq process, it gets stuck on the Calendar step and will not finish the synq process. I have to either wait for it to quit or force it to quit. Additionally, when I try to log a new event or meeting onto the Calendar on the iPhone, it will lock up and quit the app. I then have to shut down the iPhone to get it to open again but it just locks up again when I try to log a new event. Any suggestions? I'm tempted to do a back up and reinstall but don't want to risk any data loss.
