How can I set/change the font for a PDAnnot object?

I think this question is about the OLE interface, not plug-ins? If so, you cannot. It's a very old interface.
Maybe it can be done with JavaScript, I don't know the API well enough. But note that the great majority of annotation types do not accept a font; you need to understand exactly what kind of annotation you need (probably with reference to the PDF specification).
Sounds like a job for a plug-in, but working with custom fonts and annotations even there could turn out to be pretty challenging.

    @Renzo – in Acrobat Pro you have an option to format the form fields.
    Under the tab "Appearance" there is "Text" with "Font Size:", "Font:" and "Text Color:".

    You can save the settings and re-use the settings.

    Most forms (99% or more) are created for simple text input, where you cannot change anything.
    The creator of the form could allow Rich Text input (which allows you to change font, text size, color, etc.), but frankly I have never seen such a form, and I wouldn't know how they look.  But I'm sure they would show some kind of controls to alter the text appearance.

    I downloaded Tinkertool.  Found the font section and increased the size of most selections.  The only thing that changed that I can see is the window label.  The apple line, bookmarks line, the tool bar in safari are all the same---font size of about 4.  Found the finder "show view options" but that didn't seem to change anything either.  I'll keep working.  Restarted all apps after changes as well.

    Editing the source is a bit of a pain because you'll have to redo it after every update.  It's better to write yourself a mini-extension that adds a new stylesheet -- that is also very simple to do.  Just follow these instructions: https://github.com/adobe/brackets/wiki/Customize-Your-Code-Font.

    There is no way that I know of within the native iPad email app. You might want to search the app store for a third party app that lets you change fonts if this is important.
