I know this has been posted several times but I'm obviously a luddite cos I still can't do this basic task and it's driving me mad! Please someone help!
You need to read the iPhone User Guide. A link to it can be found in the Safari App on your phone or you can search Apple's website.

You can do it like KiltedTim suggested or if you are on iOS 5 you can use photostream too. I use photostream on my macbook air to import photos I take with my iphone. I never have to sync the phone up like I used to.

Just as photos that were taken more than 30 days ago remain on your MacBook Air, so will photos that are deleted from your iPhone.
Photos in My Photo Stream are saved on the iCloud server for 30 days.
Because your Mac and PC have more storage than your iOS devices, you can choose to download all of your My Photo Stream photos automatically.
In iPhoto or Aperture preferences on your Mac, select Photos (or Photo Stream) > My Photo Stream > Automatic Import. All of your photo stream photos will import into your Events, Projects, Photos, Faces, and Places folders in iPhoto or Aperture.

The laptop I was using to upload photos from my iphone is no longer good and I want to use my desktop instead but it is displaying an error that the iphone has no photos in it. How do I start using my desktop to upload my photos from my iphone5, is there any software I need to download?
Photos on the phone in the DCIM folder are those located in the camera roll. Photos that are synced to the phone from the computer cannot be accessed in this manner. They are synced and the only way to manage them is to connect the phone, go to the photos tab and un-select the folders you want to remove or go into the folder photos are located in and remove the photos you no longer want to sync. Then sync the phone and the photos will be removed.

I have this problem too. Photo stream works fine on iPhone+iPad, but the photos are never loaded to my PC (Win 8.1). My shared folders works fine on iOS and PC, but not My photo stram.
In the beginning, a couple of years ago, it work, but not now (2014).
All iOS are updated, and PC-iCloud software/panel is Active, and Photo stream is activated.
What to do??

Eric, thanks for the info about the pictures, it worked. The mail icon is not the one that came with the mac(it looks like a stamp with an eagle) this appears at a notepad with a ruler,paintbrush and pencil. I can not open it, I did not download it and now I can not turn off the computer with shutdown I have to manually turn if off by pressing the power button. This can not be normal. How can I transfer photos from an IPhone 5 to an IPad 2 using a cable. I have tried using the lightning to firewire adapter with my current lead but this only seems to allow a download from the IPad to the IPhone and not the other way around.
The devices are not designed for transfer of that kind. Use Photo Stream as suggested by another poster, or transfer photos to your computer (a good idea anyway since they will be lost if your device needs to be reset), then use iTunes to sync them to the other devices.

Pics taken with the iphone are imported to your computer as with any other digital camera ( as explained in the manual).
Pics synced to your iphone from your computer should still be on the computer.
