When restoring an iPhone with iTunes, the iPhone is completely erased first followed by the current iOS version being installed or re-installed. Following iOS being reinstalled, you are prompted to restore from your iPhone's backup which is selected by default, or as a new iPhone.
Photos/videos in the Camera Roll are included with your iPhone's backup, but if there is a problem with your iPhone's backup in regards to this data, you could lose the data and there is no way to know if there is a problem until too late.
Photos/videos in the Camera Roll can and should be imported by your computer as with any other digital camera. If not and your iPhone were lost or stolen or damaged beyond repair and not replaced for whatever reason, this data would be lost.
You should import the photos/videos in the Camera Roll by your computer on a regular basis and especially immediately before restoring your iPhone with iTunes and before installing an iOS update.
