Your i-device was not designed for unique storage of your media. It is not a backup device and media transfer was designed for you maintaining a master copy of your media on a computer which is itself properly backed up against loss. Syncing is one way, computer to device, updating the device content to the content on the computer, not updating or restoring content on a computer. The exception is iTunes Store purchased content.

You need at least two full sets of your music, not including what is on the iPod:
-- One full set on the PC within iTunes (on internal HD or ExHD)
-- One full set on an separate external backup medium (CD/DVD/ExHD/other)
-- One (full or partial) set on the iPod
Music files on just the iPod and only one other medium is not considered having any backup.
If you cannot store your complete music files on the computer’s internal HD, then create and maintain at least two external sets (any multiple combinations of ExHDs, DVDs, CDs).
Backup both the music files and the Library database file (iTunes Library.itl). What are the iTunes Library files?

iTunes purchased content on the device can be transferred to the computer, simply connect the device. Do not Sync! Select File > Transfer Purchases.
The best solution is to use the backup of the computer to put everything back.

Help! Erased Hard Drive and can't get my library from my iPod to my HD

HELP! I recently had to erase my hard drive and start fresh. I have Mac OS X 10.3.9 cleanly installed. I now have the problem that I can't get my music library from my iPod to iTunes. Many, many hours went into transferring my CD collection into iTunes. I can't fathom going through all of that again! Any suggestions would be appreciated!

I had the same problem and after installing my OS new i was able to copy back all the songs from my iPod to iTunes. Wasn't able though to keep the playlists, but maybe you find a way how to copy the songs including playlists.

No. Music only syncs one way - computer to iPod. You would need to use a third-party utility to transfer music from the iPod to the iTunes Library such as Phone to Mac - Pod to Mac

You should be able to recover the media with the tips in this post from forum regular Zevoneer but I'm not sure what the implications are for your application settings. You can probably transfer your purchases into a newly authorised library, backup the device, recover any other media using third party tools, then restore the device from the backup. That should switch the assocation of the device to the new library but it is not something I've personally tested.
When you get it all working, make a backup!

Quite possibly, at least assuming you are based in the US. Install the latest build of iTunes, sign into the iTunes Store and click the Purchased entry in the Quck Links panel. Here in the UK I only get tabs fro Apps and Books but, if I understand correctly, you should also get a tab for Music. Click on the Not in My Library button and you should be able to download your purchases again.
