If the message is stuck on the screen then a soft-reset should clear it : press and hold both the sleep and home buttons for about 10 to 15 seconds (ignore the red slider), after which the Apple logo should appear - you won't lose any content, it's the iPad equivalent of a reboot.
You can then backup to the cloud via Settings > iCloud > Storage & Backup > Back Up Now

I am trying to set up a new iPad for Christmas and keep getting this message. "This iPad has not been backed up. Backups happen when this iPad is plugged in, locked and connected to Wifi."
You click OK and the message just comes back over and over again. You can't get to any icons. I really need some help to get this set up before Christmas!This should make that message for away.

When I truned on my iPad Mini this morning the following message awaited me, iPad Not Backed Up. This iPas hasn't been backed up. Backups happen when this iPad is plugged in, locked, and connected to Wi-Fi.
I have plugged it in, allowed the screen to be locked, and it is connected to the Wi-Fi. I am unable to slide the unlock tab and clicking the OK button in the message does nothing. Does anyone have any suggestions on how to get my iPad working again?Reset iPad

I dont know why this happens.Unfortunately, "because and error occured" doesn't give you much to go on. Some general things I would try if you haven't already:
Use Disk Utility on your mac to repair disk permissions, then reboot and try backing up again.
Use Finder to go to ~/Library/Application Support/MobileSync/, then control-click on the Backup folder and select Get Info, then check Sharing & Permissions at the bottom and make sure your username has read & write priviledges
Try closing all apps, then reset your phone: double-tap the home button, tap and hold the apps at the bottom until they wiggle, tap the minus sign on all apps to close them. Then reset your phone: hold the on/off and home buttons until you see the Apple logo (ignore the off slider that appears first), and release
Try a different usb to dock connector cable; ensure that your case isn't interfering with a good connection to the dock connector at the bottom of your phone; clean the dock connector at the bottom of your phone (e.g., with a dry toothbrush); troubleshoot your usb connection (http://support.apple.com/kb/TS1286)
I know this looks like a laundry list, but unfortunately it's hard to isolate the problem without a clear error message. If the problem persists, you might try posting in the iMac or iTunes forums for other ideas. -
Since updating my iPad 2 to IOS6 and activating my icloud for backup, my iPad has permanently locked. Message on screen is "iPad Not Backed Up". I click ok but no response. Cannot turn power off and cannot restore via my pc itunes. What are my options?
Thanks to everyone who responded and related subject helped. Holding the home button and power button simultaneously for 10-15 seconds does unlock the screen. I can now use my ipad again. However, a futher problem has arisen since updating to IOS6 and activating icloud at same time, Although not sure if its related. The problem is that when connected to my PC the itunes message appears 'Cannot read ipad'. It instructs you to restore but keeps timing out after about 1 hour. This maybe internet issue but can't understand why itunes cannot read my ipad. May need to completely erase all content & reset. Or any other suggestions why this has happened?

If you are stuck on that screen and can't get out of it, reboot the iPad.

Assuming that you have iCloud backups turned on .... go to Settings>iCloud>Storage and Backup>Backup Now. -

How do i get rid of the "iPad Not Backed Up" message?
How do I get rid of the "iPad Not Backed Up" message? A week ago this happened, I searched for help, I was told to reboot and the message would go away. It worked...for awhile. The message is back, but now I cannot even reboot/reset the ipad. The message blocks me and prevents me from using the "slide to power off".
I'm not totally sure why the screen locks up like that, but it seems to happen to a number of users. I see that question being asked several time a day.
My advice would be to backup regularly with iCloud so that the message does not appear. If you are already doing so, I can only assume that it is some low memory bug that is causing it, and that is merely a guess.
