What kind of Gift Card is it? An iTunes Gift Card will only ask for the redemption code ( that you scratch off) when you click the redeem button within iTunes.

The obvious answer I think is that maybe you are not entering the letters and numbers correctly. I almost never get it right the first time when I use a gift card.
Try entering the code again - slowly and methodically. If it still doesn't work - look here for help.

One final thought - if you are using a gift card that was purchased in another country - say like you have a Canada gift card and you live in the U.S. - that card will not work in the U.S. iTunes store.

I have a credit card on file in my I tunes account, but I have been using gift cards for years. iTunes has never hit my credit card if I had enough money on my account with the gift cards.
The money should be taken from your account, if you have sufficient funds in the account. If your purchase goes over the amount that you have in the account, the entire purchase will be put on your credit card, even if you are only pennies over the account balance.

If the card has been redeemed then its remaining amount should be on your account, the card itself is now effectively zero. My account's balance shows at the top left of the store's page on my computer's iTunes, and by logging in to view my account via the Store > View Account menu option. On my devices it shows at the bottom of, for example, the Featured tab in the App Store app under my account id (or you can view your account via Settings > iTunes & App Stores and it should show on the screen that you are taken to). If your account's balance is zero then it might not show at all.

Can you provide more detail than just "doesn't work"? What happens when you attempt to redeem the card?

There is no auto-fill, you have to Copy and Paste, or Find & Replace, or if each Card is a free floating Object/s then you can click on them and option shift drag copy them into place.
