Basically, the computer thinks the music is 'lost'. Follow the prompts and try to locate it. It should navigate to the files and redraw the links which should make that pop up go away

I'm finding that songs that I have imported to Itunes are suddenly showing with "Exclamation" marks and Itunes tells me that the original file cannot be found. When I tell it to locate the file, it can't do it.

Maybe you rename it. Somehow the path to the songs have changed if not deleted.
In Finder go to "Macintosh/Users/username/music/iTunes/iTunes Media/Music" that's where all your songs located so if those songs are not there you get an exclamation mark. You can delete those entries in iTunes and re-import again.

I got a new PC and unable to get my tunes to play after I transfereed them over via a thumb drive. The message I get is, "original file cannot be found, would you like to locate" and of course it cannot locate on this PC. I no longer have that computer (gave to mother in a different state). I do have my shuffle that has all my tunes on it. I have read several things on this website from using a third party device to transfer tunes from shuffle to de-authorizing old computer and authorizing this one. No definitive instructions. Please help. I want to get these tunes to play and then buy new music. Oh and the home sharing I believe is only if you still have original computer.

The shuffle is not a backup device.
Was the media actually moved from the old computer to the new computer? If not, how did you magically make the content appear on the new system?
Use the backup of the old computer to put all the content on the new computer.

I moved my iTunes library to my external hard drive some time ago and I followed the directions listed here on the apple support page. It did copy my library and now is on my hard drive... I can open iTunes and use it to sync all of my devices but I would like to play my music... I can see all of my music files but can not play them if I click on an album or song it says the following: "(file name) could not be used because the original file cannot be found. Would you like to locate it?" I click locate but do not know where to go from there? I need a way to access all files and not to hunt down one at a time on my hard drive... I am pretty sure I deleted the original library to make space on my internal drive which was the reason for moving it. I have tried and tried to fix it and do not have a lot of time to do so with a one and two year old at my house who do not like it whenI sit down at my computer. What to do??? See Make a split library portable. Move all of the bits of the library together in the the correct shape and it should work. Or undo exactly what you did and then start over.
If you're a former Windows user don't try merging two folders together that have the same name by dropping one into the same folders as the other, as that goes badly on a Mac.

Select one of the songs in question and right click, then select "Get Info". On the main dialog page you should see where iTunes thinks the file resides. Then use Windows Explorer to see if the file is actually there. If not, find out where your music is stored.If there is not a correlation between iTunes and reality, go into iTunes and delete all of the files which are not found. Then, in iTunes, select File->Add Folder to Library and select where the music is stored. iTunes will re-add all of the music into your library.

See if you have most of the songs with 'Exclamation' symbol precede the name under your music library.
If so, double click a song with ! mark on side, you will be given an option to locate the file where the song is held. Select "locate" and direct it to your External Drive folder / file where the song is and open it.
iTunes then will give you the option to use this process to update all of your library. OK that as this will fix all automatically instead of doing them one by one.

1- on the beginning i used to purchase any App from iTunes and will be downloaded to my MBAir with out any problems .
2- But, after 2 or 3 days i will get Apps icon as inactive icon in the itunes Aplication and cannot copy them to my iPhone neither cannot open them in his iTunes and i will get an error message " selected App cannot be used because the original file cannot be found and must be re-downlaoded from the iTunes".

I just got a new computer and I transfered all of my old music via an external hard drive to the new one. All the music showed up in itunes fine so I deleted the old files. Thank you for the details of the issue you are experiencing with your purchased movie in iTunes on your iMac. It sounds like iTunes thinks the movie is on your computer, but it is not located in the iTunes Media folder.
For an issue like this, I recommend deleting the movie from iTunes and downloading it from the iTunes Store again. Use these steps to delete the movie:
Mac or PC
Launch iTunes.
From the Library pop-up menu, choose Movies.
Click Movies in the navigation bar.
Click the movie you want to delete.
Press the Delete key on your keyboard.
In iTunes 11, the movie cover art will remain visible, but a cloud icon will appear on it to allow you to download the item again (in countries that support this).

I downloaded Itunes 10 on windows vista last Saturday, but after I synched my Iphone 3gs I noticed that none of my 1000 songs where on my Iphone as well as my 55 Ringtones bought from myxer.com. Tried updating my iPhone to version 5.0.1 but error shows up: "An error occurred while backing up this iPhone [-50]. Would you like to continue updating this iPhone? Continuing will result in loss of all content on this iPhone" - which has me worried.

The first thing you need to do is (in the left side of iTunes, under devices) right click on your phone, and do a manual back up. As long as that works, this should work for you. After you back it up, go to your phone in iTunes, and do a "Restore". Click the option to restore and update. When it is done with the update, it will ask you if you want to set your phone up as a new phone, or restore from the last back up. Restore it, and after another restart, and a sync, you will have your updated phone without loosing your information.
