Yes.  iTunes will report that your photos are now being accessed from iCloud Photo Library and will no longer sync photo and videos from your computer.  If you have a Mac, you'll want to avoid enabling it until OS X Yosemite in introduced along with it's built-in photo app, which will have to ability to access iCloud Photo Library.  (Note: the Photos app may not be introduced concurrently with Yosemite, but is supposed to follow "early next year".)

    Are you asking about iCloud Photo Library (Beta) as described in: iCloud Photo Library beta FAQ - Apple Support   ?
    In that case you will still see all photos on your iPhone, even after uploading the Photos to the iCloud Photo Library.  The photos on your iPhone may be light-weight optimized previews, if you have the setting
    Settings > iCloud > Photos or Settings > Photos & Camera > iCloud Photo Library > Optimize [device] Storage   enabled.
    But as long as you have enough free storage, will the your iPhone keep the full resolution versions of the photos you access most frequently.

    No, there are no gurantees. The capabilitty exists to retrive deleted data from an iPhone, but like many things in life it is not 100% successful all the time. Be careful, there are a lot of scam artists out there. They take your money & retrive nothing. This guy is one of the best in the business, but he does not work for people in the private sector.     As you are clearly aware, it is important for you to have backups of your important data, including all of these photos you are currently keeping on your iPhone. While it is possible to use iTunes or iCloud to do backups of your iPhone, the problem becomes how to access those backups if you have deleted the photos from your iPhone. Once you have done the backup and then removed the photos from the iPhone, the only way to access the photos is by restoring the backups to the phone.     If you want to easily access the photos once they are off of the iPhone, another approach to this would be to import your photos from the iPhone into an application on your Mac, such as iPhoto. To learn how to do this, use the steps in this article -

    If the photos were in your camera roll when you last backed up, restoring the backup should recover them.  Restoring an iCloud backup requires first erasing your phone.  If you have any newer data on your phone since the backup that you want to keep, you'll need to save that first.  Then go to Settings>General>Reset, tap Erase All Content and Settings, go through the setup screens on the phone and when given the option, choose Restore from iCloud Backup.  Sign into your iCloud account and choose your last backup to restore to.
    Make sure you phone is connected to wifi and your charger as this may take some time to finish.

    If you use your computer to put a picture/video on the phone, you will have to use your computer to remove it.

    There is no syncing process to move synced photos from an iOS device back to a computer.
    If you have only a small amount of photos you can email them to yourself.
    There are third party applications available online, some free and some at cost that will pull pictures off your device and back to the computer if your number of photos is great.

    If you've synced photos to your iPhone through iTunes, change your sync settings in iTunes to not sync any photos and apply the change.
    That will remove all synced photos, but not photos that you've taken with your iPhone's camera -- for those if you've imported them to your computer then to remove them you have to delete them one by one from the iPhone, or use your computer (assuming you have a Mac, iPhoto should allow you to select and delete what's stored on the iPhone, except for synced photos via iTunes).
    Then there's iCloud photos.  iCloud only holds the last 30 days of photos, but your device will continue to hold them up to the 1000 photo limit, regardless of how old they are.  To reduce these, you can delete one by one, or if you Restore your iPhone that will strip off all iCloud photos, and add back only the last 30 days of iCloud photos, assuming you leave the Photo Stream setting ON in iCloud settings.

    You can stop new photos being automatically added to Photostream in iPhoto from iPhoto preferences > Photostream.

    HOWEVER a best practice to is to never have any computer program (including iPhoto) delete the photos from the card but to import the photos and keep them and then after at least one successful backup cycle has completed and then reformat the card --  I use three very large (32 GB) cards in rotation so I do not reformat for typically a year or more giving me one more long term backup of my photos
    If you continue letting the computer delete your photos you will lose them sooner or later

    If you have iCloud and shared Photo Stream turned on, you can go into your each of your albums and do the following:
    1.     Tap on Edit
    2.     Tap on each photo you want to save back to your computer
    3.     Tap on Share
    4.     Tap on Photo Stream
    5.     Tap on the unnamed Photo Stream folder just created
    6.     Type in your iCloud or Apple ID email address
    7.     Name the Photo Stream
    8.     Tap on Next to create the shared Photo Stream album
    9.     Tap on Post
    10.   Tap on the new Photo Stream album
    11.    Tap on Edit
    12.    Tap on the photos to select them
    13.    Tap on Share
    14.    Tap on Save to Camera Roll
    Once they are back in the Camera Roll, you should be able to import them.

    If you cannot get them by restoring from backup then they are gone.
    Why would you update anyhting without making sure that all of your pics were on your computer and backed up?
    You should never update/reinstall any software without making sure that everything is backed up.
